Photo provided</figcaption> </figure> <p>Associate Professor <a href="https://volgenau.gmu.edu/profiles/rvenezia">Remi Veneziano</a> and Professor <a href="/profiles/pchitnis-0">Parag Chitnis</a> in the <a href="https://bioengineering.gmu.edu/">Department of Bioengineering</a> received a Tier 2 VIPC award to advance the development of a contrast agent for photoacoustic imaging. The two-year, $200,000 grant will support efforts to scale-up production and demonstrate that the material can be manufactured consistently for translation outside of a laboratory setting.</p> <p>If successful, the work could provide a standardized, scalable contrast agent for photoacoustic imaging and expand its use across biomedical research.</p> <p>Photoacoustic imaging combines light and ultrasound to visualize biological tissues. While the technique can detect naturally occurring molecules such as hemoglobin, it lacks widely available contrast agents that can target specific tissues or disease sites. As a result, researchers often create their own materials, which vary from lab to lab.</p> <p>Veneziano <a href="https://www.sciencedirect.com/science/article/pii/S2213597922001021?via%3Dihub">began working on the problem about eight years ago</a> with collaborators at 911±¬ÁĎ. His team focused on developing a contrast agent using indocyanine green, an FDA-approved dye. Instead of relying on complex chemical processes, the group created a method based on self-assembly, allowing the dye molecules to form aggregates with controlled size and surface properties.&nbsp;</p> <p>“We figured out a method to control the size of the assembly without the need for complex chemistry,” Veneziano said. “When you reduce the chemicals and the number of steps, you make it easier for manufacturing and future approval or at least to make something safer.”</p> <p>The team refined the process to the point where researchers in the lab can reliably produce aggregates with specific characteristics tailored for specific applications. Early funding, including a previous VIPC Tier 1 award and a university seed grant, supported initial development and validation. The group published its findings in 2023 and began building collaborations with other researchers.</p> <p>A key step in the work involved combining standard indocyanine green dye molecules with modified versions of this dye that allow for additional functionality, such as targeting specific cells or adding other fluorescent molecules. That approach helped turn the material into a more flexible platform that can be adapted for different research needs.</p> <p>The current grant focuses on what Veneziano described as the transition from a “lab product” to something that could be used more broadly. That includes demonstrating reproducibility across batches, establishing quality control standards, and determining how long the material can be stored without losing effectiveness.</p> <p>“When you want to commercialize a product, you need to demonstrate reproducibility and quality control,” he said. “From one batch to another batch, you always want the same properties.”</p> <p>Veneziano and his collaborators are also working with <a href="https://ott.gmu.edu/">911±¬ÁĎ’s Office of Technology Transfer</a> and filed a patent application related to the technology. The team founded a start-up, NanOptical Biomedical Inc., to pursue commercialization, and the team plans to seek additional funding through the Small Business Innovation Research and Small Business Technology Transfer programs.</p> <p>At the same time, the research continues to evolve. Our research is focused on using the tools of micro- and nanotechnology for applications in biomaterials and tissue engineering. The goal of current research is to design implants that induce controlled, guided, and rapid healing in cardiovascular implants such as stents and heart valves; and orthopedic implants such as hip and knee joint implants. In addition to the acceleration of normal wound healing phenomena, these implants should result in the formation of a characteristic interfacial layer with adequate biomechanical properties.</p> <p>To achieve these goals, however, a better understanding of events at the tissue-material interface is needed, as well as the development of new materials and approaches that promote biointegration. Our work proposes the use of well-controlled nanostructured interfaces to enhance implant integration. We hypothesize that controlled biomimetic nanoscale architectures can promote cell differentiation and matrix production and enhance short-term and long-term integration. Moreover, the ability to create model nano-dimensional constructs that mimic physiological systems can aid in studying complex tissue interactions in terms of cell communication, response to matrix geometry, and effect of external chemical stimuli. As a student, his&nbsp;interest in neuroscience and immunology led him&nbsp;to pursue his diploma thesis on studying microglia and their role in Alzheimer’s disease. Dannenberg&nbsp;continued this inquiry in his PhD&nbsp;thesis in Experimental Epileptology and Cognition Research in Bonn, Germany studying neuronal ensembles, neural engrams, and their modulation by acetylcholine in-vivo. After finishing his&nbsp;PhD&nbsp;thesis, he&nbsp;joined the laboratory of Michael E. Hasselmo in Boston, Massachusetts,&nbsp;where he continued experimental and computational work on the cholinergic role in memory. In 2021, Dannenberg&nbsp;joined the Department&nbsp;of Bioengineering and Interdisciplinary Program in Neuroscience as an assistant professor. Veneziano is currently focusing his research on designing and synthesizing new composite nanomaterials, combining structured 3D DNA nanoparticles with proteins, and lipids. His lab is using these constructs to investigate fundamental questions about the role of biomacromolecule nanoscale organization in cell membrane interaction events.</p> <p>Veneziano was previously a postdoctoral associate in the Department of Biological Engineering at MIT under the supervision of Prof. Mark Bathe where he made important contributions in the field of DNA nanotechnology. He notably participated in the development of a new class of 3D DNA nanostructures for biomedical applications and developed a method for the large-scale production of long single-stranded DNA scaffolds. Before this, she was a professor in the Chemical Engineering Department at the Ecole Polytechnique in Montreal, where she spent 15 years directing a federally-funded research program on the role of inflammation in articular cartilage repair. She also brings five years’ experience as a research director at a Montreal-based biomedical device company where she invented and developed a medical device for articular cartilage repair that achieved regulatory approval for use in over 17 countries, and was eventually acquired by Smith &amp; Nephew.</p> <p>Professor Hoemann is highly regarded internationally for her work on cartilage and bone tissue engineering and biomaterial-induced blood and innate immune responses. She is the recipient of 2 NIH-Fogarty post-doctoral fellowships, and four career fellowships, is a fellow member of the International Cartilage Repair Society, and serves on the editorial boards of Cartilage and The Open Orthopaedics Journal. She is a co-founder and on the board of directors of ORTHO-RTi, an orthopedic biotech company specializing in implants that repair joint tissues. Her research program focuses on understanding how to use biomaterial-guided immune responses to regenerate bone and cartilage tissues. She has published 68 peer-reviewed papers, 14 book chapters/expert opinion papers, 171 conference abstracts, and 8 patent inventions. Her current research involves studying bladder neurophysiology and working on a closed-loop neuroprosthesis for bladder control. In general, Ross’ research interests are in the areas of neural engineering and neuromodulation. The lab’s research focuses on developing and applying computational methods to advance the diagnosis and treatment of disorders of the oculomotor system and the musculoskeletal system. Using clinical data-driven biomechanical models, ocular ultrasound imaging, and a&nbsp;robotic platform of the ocular motor plant, the lab examines the coordination of extraocular muscles as well as the biomechanics of strabismus. Applying quantitative ultrasound image analysis of the pelvic floor, the lab studies childbirth-related pelvic floor muscle injuries. After attending universities in Brazil (Unicamp and USP) and Germany (Stuttgart and Bonn), she was a researcher at Stanford. She still likes to travel and her group collaborates with universities in Israel, Ireland, Peru, and Korea.</p> <p>The <a href="https://neural.bioengineering.gmu.edu" target="_blank" title="Neural Engineering Lab website">Neural Engineering Lab</a> is affiliated with the ECE and Bioengineering Departments. Our team has interests in Chemistry, Math, Physics, and several Engineering disciplines. High school, undergraduate, and graduate students perform team-based projects on assistive technology, wearable embedded systems, neural implants, and bioinstrumentation.</p> <p>Nathalia’s activities center around teaching Circuits, Team-Based projects, and Neural Prosthetics (graduate-level class). She attends (too many) meetings but she would rather stay in the lab and play with electronics, soldering, and cell cultures. <p>Siddhartha Sikdar is a distinguished university professor in the Department of Bioengineering at 911±¬ÁĎ. He is the co-director of the <a href="https://medtech.gmu.edu/" title="CMI">Center for MedTech Innovation (CMI)</a>. This transdisciplinary center includes faculty with expertise in engineering, neuroscience, and rehabilitation. The vision for CMI is to improve the function and quality of life of individuals living with physical and psychosocial disabilities through research on mechanisms and modulation of complex adaptive systems underlying human movement and behavior.</p> <p>Sikdar’s research group within CMI conducts translational research using imaging to investigate brain-body interactions in several clinical conditions of major public health significance, such as chronic pain, stroke, spinal cord injury, and amputation. The group uses state-of-the-art ultrasound and laser instrumentation for developing new ultrasound, optical, and hybrid imaging techniques, and assistive technologies. The group is currently investigating the novel use of wearable ultrasound sensors for sensing muscle activity and controlling external devices such as multi-articulated prosthetic hands and exoskeletons.</p> <p>Sikdar earned his PhD in Electrical Engineering from the University of Washington, Seattle in 2005. He received a postdoctoral fellowship from the American Heart Association. He has been a recipient of the NSF CAREER Award, the Volgenau School of Engineering Rising Star Award, Mason’s Emerging Researcher/Scholar/Creator Award and was nominated for the Rising Star Faculty Award of the State Council of Higher Education in Virginia. His current research is funded by the National Science Foundation, the National Institutes of Health, the Department of Defense, and the Veteran's Administration.</p> <figure class="quote"> <p><strong>Research Spotlight:</strong></p> <div> <p>​Bioengineering professor Siddhartha Sikdar, co-director of the Center for MedTech Innovation (CMI), uses cutting-edge ultrasound technology to help people gain greater control of prosthetic devices for their arms, hands, and legs. The successful completion of this research will lead to the first human evaluation of an integrated prototype that uses low-power portable imaging sensors and real-time image analysis to sense residual muscle activity for prosthetic control.
